I have Mediaface 5.0.40. How do I print on the Adhesive Spine (CD Plus Sheets)? I can't get the print to go longwise. If I print one letter at a time I can get the label, but I want to rotate it 90 degrees.

There is no instruction on how to configure MediFACE to print as you mentioned in your question, but I've found a printing guide/user manual that you can check to see some different printing methods.
You can download the PDF file using the following link. You'll need a PDF reader to be able to open the file.
I have downloaded Windows 10 and can't open MediaFACE 5 (advanced) on my computer.

The software isn't available anymore, therefore there is no Windows 10 compatibility. The official website called mediaface.com now redirects to another website that's based on the MediaFace application.
You can use a solution from the SmileSoftware called DiscLabel. The application is compatible with OSX 10.6 or later. Use the URL below to access its official website where you can download a trial version of the software.
I am trying create two different labels in 2 up page so that I will be able to print one time. I could duplicate the label, but can not edit the 2nd label. Is there a way this can be done?

The first thing I would recommend doing is checking the built-in Help section. If the software doesn't have that section, locate the installation folder and look for Readme files or User Guides in PDF or CHM format. However, I have installed the application and used a template from the available list. I was able to print it, but the software doesn't allow the operation you have mentioned in your question. Try creating 2 labels and then export them to another format (eg. as image) and load Photoshop or Gimp and add them to one page only. Before you do that, apply all the edits to both labels and then export.

You can start the designer using the start page: http://www.design.neato.com/
Click on an item and then wait for the application to load properly.

Additionally, check the database of Mac Informer for alternative software, in case you don't like DiscLabel.
